Pestisit klorprifosun rat (wistar albino) karaciğerinde oluşturduğu hasar üzerine kurkuminin antioksidan etkisinin incelenmesi

Öz

Bu
çalışmada dünyada yaygın olarak kullanılan geniş spektrumlu pestisit olan
klorprifosun (CPF) ratların (Wistar albino) karaciğerinde oluşturduğu hasarın
miktarı ve bu hasarın giderilmesinde antioksidan özelliği bilinen kurkuminin
koruyucu rolünün incelenmesi amaçlanmıştır. Bu amaçla, bir kontrol grubu ile
sırasıyla sadece CPF, sadece kurkumin ve CPF+kurkumin verilen 3 muamele grubu
olmak üzere toplam dört grup oluşturulmuştur. Her bir grupta 12 adet 90 günlük
erkek rat kullanılmıştır. 5 mg/kg/gün CPF ve 100 mg/kg/gün kurkumin gavaj
yoluyla hayvanlara verilmiştir. 15. ve 30. günlerde gruplardaki 6’şar hayvan
sakrifiye edilmiştir. Histolojik takip işlemlerinden sonra ışık mikroskobu ile
yapılan değerlendirmede CPF grubunda kontrol grubuyla karşılaştırıldığında
karaciğerde hidropik dejenerasyon, inflamasyon odakları, sinüzoidlerde
genişlemeler, hepatositlerde sitoplazmik değişiklikler, glikojen yoğunluğunda
farklılıklar gözlenmiştir. CPF ile birlikte kurkumin verilen grupta, CPF
grubuna göre daha az oranda histolopatolojik değişiklik gözlenmiştir. Elde
edilen bulgular kurkuminin, CPF’nin neden olduğu histopatolojik hasarı
indirgediğini göstermektedir.

Investigation of curcumin’s antioxidant effect on pesticide chlorpyrifos caused damage in rat (Wistar albino) liver

Öz

In this study, broad-spectrum pesticide chlorpyrifos (CPF)
caused damage on rat (Wistar albino) liver and protective role of curcumin, a
well known antioxidant were investigated. For this purpose, 4 groups were
created, including a control group with 3 treatment groups; only CPF, only
curcumin and
CPF + curcumin,
respectively. Each group was consisted of 12 male rats. 5 mg/kg/day CPF and 100
mg/kg/day curcumin were given to the animals via gavage. 6 animals in each
group were sacrificed in 15th and 30th days. Following histologic examinations,
when the CPF group compared with the control group; hydropic degenerations,
focal inflammatory cell infiltration, expansion in the sinusoidal, cytoplasmic
changes in hepatocytes and glycogen concentration differences were observed by
light microscopic evaluations. It was observed that CPF + curcumin-treated
group had histological changes than CPF group. The results showed that curcumin
has a reduction effects on CPF induced histopathological damage.
