Effect of Iturin A on Oxidative Stress and Immune Response in Breast Cancer Cell Lines

Öz

This study aimed to evaluate the effects of Iturin A on oxidative stress and DNA damage, and immune response in MCF-7 and MDA-MB-231 breast cancer cells. Total antioxidant capacity (TAC), total oxidant status (TOS), and 8-hydroxy-2′-deoxiguanosine (8-OHdG) levels were determined by spectrophotometric and ELISA methods, oxidative stress index (OSI) was calculated. NF-κB and COX-2 gene expression levels were analyzed using RT-qPCR. Iturin A significantly increased TAC (p<0.0001 in both), TOS (p<0.0001 in both), and 8-OHdG (p<0.01 and p<0.001, respectively) levels in both MCF-7 and MDA-MB-231 cells. Iturin A treatment significantly increased OSI levels in MCF-7 cells (p<0.001), although an increase in MDA-MB-231 cells was not statistically significant (p>0.05). Iturin A treatment significantly reduced NF-κB expression in MCF-7 cells (p<0.0001) and MDA-MB-231 (p<0.05) cells. But, the observed changes in COX-2 gene expression were not statistically significant in either cell line (p>0.05). Iturin A may differentially modulate oxidative balance and immune response in estrogen receptor-positive and -negative breast cancer cells. These findings suggest that Iturin A could be a potential agent targeting redox and inflammation balance in breast cancer therapy.
