Bracon didemie Beyarslan (Hymenoptera, Braconidae)’nin Ephestia kuehniella (Zeller) ve Galleria mellonella (L.) (Lepidoptera: Pyralidae) üzerinde kültür yöntemi

Öz

Tuta absoluta (Meyrick) (Lepidoptera: Gelechiidae) parazitoiti, Bracon didemie Beyarslan (Hymenoptera: Braconidae)’nin Ephestia kuehniella (Zeller) ve Galleria mellonella (L.) (Lepidoptera: Pyralidae) üzerinde kültür yöntemi tanımlanmıştır. Bu parazitoite konukçu olarak E. kuehniella’nın irmikte üretilmiş son dönem; G. mellonella’nın ise özel larva besi karışımında üretilmiş 3-4. dönem larvaları kullanılmıştır. Çalışmalar 25±1 oC ve % 70±5 orantılı neme ayarlı iklim dolaplarında yürütülmüştür. E. kuehniella’nın 50’şer larvası 17x12x6 cm boyutlarındaki plastik kapların tabanına 0.3 cm kalınlığındaki irmik tabakası üzerine; G. mellonella larvaları ise 12 cm çap ve 23 cm yüksekliğindeki silindirik plastik kapların tabanına 0.5 cm kalınlığındaki yukarıda belirtilen besi karışımı üzerine 20 adet bırakılmıştır. Plastik kapların iç yüzeylerine erginlerin beslenmesi için bal damlacıkları sürülmüştür. Kültürler E. kuhniella’da 10; G. mellonella’da 3 tekerrürlü olarak yürütülmüştür. Ephestia kuehniella kültüründe B. didemie bir dölde 4,62 kat popülâsyon artışı göstermiş (ortalama 45.20±5.63 ergin/50 larva/33,8±3,46 gün), üretim 6 döl boyunca sürdürülmüştür. Galleria mellonella kültüründe ise, bir dölde 2,71 kat popülasyon artışı (ortalama 25,33± 4,07 ergin/20 larva/25,67±1,28 gün) gözlenmiş; üretim dört döl sürdürülmüştür. Çalışmalarda G. mellonella larvalarının kokon örerek kendilerini parazitlenmekten korudukları görülmüştür.

Culture methods of Bracon didemie Beyarslan (Hymenoptera: Braconidae), parasitoid of Tuta absoluta (Meyrick), on Ephestia kuehniella (Zeller) and Galleria mellonella (L.) (Lepidoptera: Pyralidae) were described. Last instar larvae of E. kuehniella and 3rd-4th instars larvae of G. mellonella were used as hosts at 25±1 oC and 70±5 RH%, in dark conditions. Fifty larvae of E. kuehniella in plastic containers (17x12x6 cm) with a 0.3 cm layer of semolina and 20 larvae of G. mellonella in plastic jars (12 cm dia., 23 cm high) with 0.5 cm larval diet were placed. Honey droplets were put on inner surfaces of the containers as adult diet. One to two days old 6-18 adult parasitoids, were released into the containers. Cultures have 10 replicates for E. kuehniella and 3 for G. mellonella. Population increase of B. didemie was 4.62 fold per generation in E. kuehniella cultures (mean 45.20 ± 5.63 adult/50 larvae/33.8 ±3.46 days), maintained for 6 generations. In G. mellonella cultures, maintained for 4 generations, the population increase was 2.71 fold per generation (mean 25.33± 4.07 adults/20; larvae/25.67±1.28 days). Larvae of G. mellonella were observed to produce cocoons to protect themselves from being parasitized.
