Sıçanlarda Pilocarpin ile Oluşturulan Epilepsi Modelinde Topiramat'ın Antikonvulsan Etkileri

Anticonvulsan Effects of Topiramate in the Pilocarpine Model of Epilepsy in Rats

Öz

The effects of topiramate (TPM) therapy on zinc translocation and anticonvulsan outcomes have been researched in this study. Also whether TPM has a role on antiepileptic effect by affecting the zinc ions on the hippocampus has been investigated. Adult, male Spraque-Dawley rats between 300-350g. have been used in the experiments. To receive the electrocorticographic (ECoG) data from the rats, 5 stainless steel screw electrodes were implanted epidurally under anesthesia (thiopental sodium, 40 mg/kg, i.v.) to all of the rats and canuls were placed in the left lateral ventricles for i.c.v. injection before the experiments. Seizures were provoked by pilokarpine-HCl injection (380 mg/kg i.p.). Ten minutes before pilocarpine-HCI injection; 20, 60 and 100 mg/kg topiramate (i.p.), CaEDTA (100 mM i.c.v.) and ZnCl2 (35 mg/kg i.p.) have been introduced into the groups accordingly. Concurrently, the behavioral variances of the animals were observed during this process. After two hours of pilocarpine injection, seizure were terminated by diazepam injection. By the end of 24 hours, the rats under anesthesia were decapitated, their brains were removed rapidly and the state of zinc ions was analyzed by hippocampal staining (TSQ). When the status epilepticus (SE) incidence, 24-hour survival, and the seizure scores were analyzed, the anticonvulsan effects of TPM were confirmed. The amplitude and frequency of the spikes in the ECoG records were less in all of the TPM groups. When 100 mg/kg was applied, TPM was effective on the zinc translocation. This shows that TPM prevents the translocation of zinc ions from the presynaptic terminals of mossy fibers to postsynaptic neurons.
