Double Skeletal Staining With Microwave Irradiation

The aim of this study is to shorten the 48-72 hours lasting (48-72 hours-lasting) staining period of the most frequently used double skeletal staining method, Inouye's method, by microwave irradiation. 24 one-day-old Swiss albino mice were separated into two groups (microwave group and control group). Cases in the microwave group were stained by microwave oven at 40.47±4.8 °C for 8 hours and cases in the control group were stained by conventional heating at 37 °C for 72 hours, as described by Inouye. When the staining quality of the cases compared no significant difference was found between groups. By the use of microwave irradiation, the duration of staining procedure, which originally was 72 hours, has been reduced to 8 hours without causing any reduction in the quality of staining. It is concluded that microwave irradiation can be used to decrease the time required for double skeletal staining.
