Uçucu Yağ ve Arbusküler Mikorhizal Fungus’un Domates Kök ve Kök Boğazı Çürüklüğü (Fusarium oxysporum f. sp. radicis lycopersici Jarvis & Shoemaker) Hastalığına Etkileri

Abstract

Bu çalışmada, bazı Arbusküler Mikorhizal Fungus (AMF) türleri (Rhizophagus intraradices (Ri) ve Funneliformis mosseae (Fm)) ile bazı uçucu yağların (kekik, nane ve adaçayı), domatesteki önemli hastalıklar arasında yer alan Fusarium oxysporum f. sp. radicis- lycopersici (Jarvis & Shoemaker) (FORL)‘nin yol açtığı kök ve kök boğazı çürüklüğü hastalığı üzerine etkileri incelenmiştir. Çalışmanın birinci aşamasında, in-vitro koşularda üç farklı uçucu yağ ile bunların beş farklı dozu (25, 50, 75, 100 ve 150 μl L-1), FORL’ye karşı denenmiştir. Çalışma sonucunda patojenin miseliyal koloni gelişiminin sadece kekikten elde edilen uçucu yağın (KUY) tüm dozlarının %50.8-80.8 oranında engellediği; en iyi dozun ise 150 μl L-1 doz olduğu saptanmıştır. Çalışmanın ikinci aşamasında in-vivo koşullarında yetiştirilen domates bitkilerine, AMF türleri ile 150 μl L-1 dozundaki KUY uygulanmıştır. Uygulama gruplarının bitki gelişimine önemli bir etkileri olmamakla beraber, AMF kolonizasyon yoğunluklarına ve mikorizal bağımlılık kriterlerine göre en iyi uyumun Ri ile olduğu saptanmıştır. Çalışmanın üçüncü aşamasında Ri ile KUY’un 150 μl L-1 doz uygulamasının, domates bitkisinde FORL’ye olan etkileri araştırılmıştır. Tüm uygulamalar kontrol grubuna göre hastalığı %28-55 oranlarında baskılarken, Ri+KUY+FORL kombinasyonun patojene karşı en etkili grup olduğu, uygulama gruplarının bitkinin morfolojik gelişimine önemli katkı sağlamadığı gözlenmiştir. AMF kök kolonizasyon yoğunluğu açısından en yüksek değerler Ri ve Ri+KUY+FORL uygulama gruplarında sırasıyla, %43.89 ile %37.77 oranlarında olurken, AMF spor yoğunluklarında istatistiki olarak önemli fark oluşmamıştır. Mikorizal bağımlılık ise sadece Ri+KUY uygulamasında tespit edilmiştir. Sonuç olarak, bu çalışmada kullanılan Ri ile KUY uygulamalarının patojeni engellemede önemli bir potansiyele sahip olduğu, ancak bitki gelişimine önemli katkı sağlamadığı belirlenmiştir.

The Effects of Essential Oils and Arbuscular Mycorrhizal Fungi on Decay Disease (Fusarium oxysporum f. sp. radicis lycopersici Jarvis & Shoemaker) of Root and Root Collar of Tomato

Abstract

In this study, the effects of some Arbuscular Mycorrhizal Fungi (AMF) species (Rhizophagus intraradices (Ri) and Funneliformis mosseae (Fm)) and some essential oils (thyme, mint and sage) on Fusarium oxysporum f. sp. radicis lycopersici (Jarvis & Shoemaker) (FORL) of root and root collar of tomato which are two of the important diseases were invesitigated. In the first stage of study three different essential oils and five different doses of oils (25, 50, 75, 100, 150 μl L-1) have been tested against FORL in-vitro conditions. As a result of the study, it was determined that all doses of essential oils obtained from thyme (KUY) prevented the development of micellar colonies of the pathogen by 50.8-80.8%; and the best dose was 150 μl L-1. In the second stage of the study, 150 μl L-1 dose of KUY was applied to tomato plants grown under in-vivo conditions with AMF species. Although treatment groups did not have a significant effect on plant growth, according to AMF colonization densities and mycorrhizal dependence criteria, the best effect was found with Ri. In the third stage of the study, the effects of 150 μl L-1 dose of KUY and Ri application on FORL in tomato plant were investigated. Ri+KUY+FORL combination was determined to be the most effective group against the pathogen. In addition, although all treatments suppressed the pathogen at a rate of 28-55%, they did not show significant effect on the morphological development of the plant. While the AMF root colonization density Ri and Ri+KUY+FORL treatment groups had the highest values at 43.89% and 37.77% respectively, there was no significant difference in spore density in the rhizosphere Mycorrhizal dependence was detected only in Ri+KUY treatment. As a result, it was determined that Ri and KUY treatments have an important potential in preventing the pathogen, but they do not contribute significantly to plant growth.
