STR HUM THO1’ in PCR Ürünlerinin Kapiller Jel Elektroforezi

Öz

HUM THO1 sisteminin STR lokusu Adli Tıp araştırmaları için hızlı, kolay, güvenilir bilgi alınabilen polimorfik DNA lokuslarındandır. Bu sistemler, PCR sayesinde nesep tayini ve Adli olayların aydınlatılmasında son derece umut verici bilgiler sunmaktadırlar. İnsan kaynaklı genomik DNA kelatlaştırma (Chelex 100 ) yöntemiyle izole edildi. STR lokusunun çoğaltılması için Gene Print STR kiti (Promega Corporation,USA), ve kitin kullanma kılavuzuna uygun olarak THO1 lokusunun PCR tekniği ile çoğaltılması sağlandı. THO1 lokusunun PCR ürünleri denatüre edilmiş %4’lük poliakrilamid jel elektroforezi ve gümüş boyama yöntemiyle analiz edildi. Tamamlanan ve tekrar edilebilir olan STR HUM THO1 alellerinden 191 ve199 baz çifti büyüklüğündeki bantları, net ve tekrarlanabilir bir şekilde gözlendi. Kapiller elektroforez elektroforeogramları denatüre edilmiş %4’lük poliakrilamid jel elektroforez bantları ile karşılaştırıldı. Bu çalışmada, kapiller jel elektroforez (CGE)'nin az miktarda örneğe gereksinim duyması, hızlı ve çabuk ayırım süresi, yüksek ayırıcılığı, kolay miktar hesabı ile otomasyona uygunluğu gibi özelliklerinden yola çıkılarak STR HUM THO1’ in PCR ürünlerinin ayırımı amaçlandı.
