Tüketime Hazır Bazı Gıdalarda Staphylococcus aureus’un Varlığı, Enterotoksijenik Özellikleri ile Antibiyotik Dirençliliğinin Belirlenmesi

Öz

Bu çalışma, Ankara’da satışa sunulan tüketime hazır bazı gıdalarda Staphylococcus aureus’un varlığı, enterotoksijenik özellikleri ile antibiyotik dirençliliklerini belirlemek amacıyla yapılmıştır. Bu amaçla her birinden 20’şer adet olmak üzere alınan (Rus salata, İtalyan salata, Tavuk salata, kadınbudu köfte, arnavut ciğer, tavuk döner, kremalı yaş pasta, kazandibi ve keşkül tatlı örneği) 180 adet örnek materyal olarak kullanılmıştır. Alınan örneklerde S. aureus’un varlığı klasik kültür tekniğine göre tespit edilmiştir. Daha sonra klasik kültür tekniğinde S. aureus olarak belirlenen suşlar PCR tekniği ile doğrulanmıştır. Doğrulanan S. aureus suşlarında PCR tekniği kullanılarak klasik enterotoksin genleri (sea, seb, sec, sed, see) ile mecA ve mecC genleri araştırılmıştır. Ayrıca S. aureus suşlarının 9 farklı antibiyotiğe karşı dirençlilikleri araştırılmıştır. Yapılan analizler sonucu toplam 13 adet S. aureus suşu izole edilmiştir. İzole edilen bu suşlardan 1’i (% 5) kadınbudu köftede, 2’si (% 10) arnavut ciğerinde, 1’i (% 5) tavuk dönerde, 1’i (% 5) tavuk salatasında, 4’ü (%20) kremalı yaş pastada, 1’i (% 5) kazandibinde ve 3’ü (%15) keşkül örneklerinde saptanmıştır. Rus salatası ve İtalyan salatası örneklerinde ise S. aureus bulunmamıştır. İzole edilen toplam 13 adet S. aureus suşundan 7’sinin (% 53,8) enterotoksijenik özellikte olduğu belirlenmiş olup, 4’ünün sea, 3’ünün ise sec enterotoksin genine sahip olduğu belirlenmiştir. Ayrıca suşların hiçbirinde mecA ve mecC geni tespit edilmemiştir. Yapılan antibiyotik dirençlilik testlerinde suşlardan 8’nin (% 61,53) penisiline, 6’sının sefoksitine (% 46,15), 4’ünün (% 30,76) oksasiline, 3’ünün (% 23,07) eritromisin ve siprofloksasine (% 23,07), 2’sinin (% 15,38) klindamisine,1 izolatında (% 7,69) trimetoprim/sulfametoksazola dirençli bulunmuştur. Suşların hiçbiri gentamisin ve kloramfenikola karşı dirençli bulunmamıştır. Sonuç olarak, tüketime hazır gıdalarda bulunan enterotoksijenik S. aureus suşlarının gıda zehirlenmelerinde önemli rol oynayabileceğini göstermektedir. Bu nedenle halk sağlığının korunması ve güvenli gıda üretiminde HACCP sisteminin etkin olarak uygulanması önemlidir.
