Kocayemiş’in (Arbutus unedo L.) Doku Kültürü İle Üretimi

Öz

Akdeniz ekosisteminin karakteristik türlerinden biri olan kocayemiş, diğer adı ile dağ çileği ülkemizde doğal yayılış gösteren türlerden birisidir. Ekonomik açıdan yetiştiricisini memnun edecek bir tür olarak ilgi beklemekte olan bu tür yeterince tanınmamaktadır. In vitro’da; sürgün ucu (3-5 mm), lateral gözler ve üzerinde genellikle bir göz bulunan nodlar olmak üzere üç tip eksplant kullanılmıştır. Sürgün ucu kültüründe kararmalar meydana gelmiştir. Yan gözlerde de kararma ve geç, cılız gelişme olduğu görülmüştür. Üzerinde genellikle bir göz bulunan nodlarda (10-15 mm) ise en iyi gelişme görülmüş ve bu eksplant tipinden gelişen mikro sürgünler ile sitokinin ve oksin doz denemeleri yapılmıştır. Köklendirme çalışmaları in vitro ve in vivo’da yapılmıştır. In vitro’da 3 ortam ve IBA’in 2 dozu kullanılmıştır. In vitro’da en yüksek köklenme %86,7 oranında K/3 tipinde, makro elementlerin yarıya indirgendiği MS ortamında ve 5 mg/L IBA dozunda oluşmuştur. En düşük köklenme ise K/1 tipinde %25 oranında MS ortamında ve 2 mg/L IBA dozunda gerçekleşmiştir. In vivo’da mikro sürgünlerin dip kısımları IBA’in 500 ve 1000 ppm dozlarına 10 saniye batırılmış ve nemli perlit içeren plastik kaplara dikilmiştir. In vivo’da ise en yüksek köklenme %58,3 oranında 1000 ppm IBA dozunda K/1 ve Esenköy tiplerinde meydana gelmiştir.

Micropropagation of Strawberry Tree (Arbutus unedo L.)

Öz

The strawberry tree (Arbutus unedo L.) is native to the Mediterranean region that also widely distributed in Turkey. Its farming can be profitable, but it is not well known enough in our country. In this study, shoot tip (3-5 mm), axillary bud and nod with usually one bud (10-15 mm) as explant were used. Severe browning was observed in shoot tip culture of A. unedo. The micro-shoots of axillary bud explants were also occurred browning, late sprouting, weak development and opening in color. The micro-shoots originated from nodes showed the best improvement and cytokine and auxin dose trials were established them. In vitro and ex vitro rooting experiments were carried out in three different media and two concentrations of IBA. The highest rooting rate with 86,7% was obtained the micro-shoots originated from K/3 type in MS medium added 5 mgl-1 IBA and ½ macro elements in vitro, while K/1 type showed the lowest rooting rate with 25% in MS medium added 2 mgl-1 IBA. 500 and 1000 ppm concentrations of IBA were used at ex vitro rooting. The micro-cuttings in height 2-3 cm were soaked in IBA solution for 10 s and then they were planted in plastic jars containing perlite. K/1 and Esenköy types had have the highest rooting rate of rooting with 58,3% in 1000 ppm IBA concentration.
