Effect of Follitropin Alpha on Matrix Metalloproteinase-7 Expression in Uterus in Rats

Öz

Follitropin alfa is a type of follicle-stimulating hormone belonging to the hormone family registered as gonadotropins and is used in assisted reproductive technology. In our study, we aimed to demonstrate the expression of Matrix Metalloproteinase-7 in the uterine tissue of animals given Follitropin alfa. In the study, 16 adult female Wistar albino rats with an average weight of 200-220 grams, 8-10 weeks old, were recorded. Follitropin alfa (4IU) filling was administered in a manner that divided the diestrus phase into three segments. Uterine tissues were taken at the end of 21 days. Tissue samples were fixed in 10% buffered neutral formalin solution. Periodic Acid-Schiff and Hematoxylin-eosin staining methods were used for general morphology in tissue uterine samples from which 3 µm sections were taken, and immunofluorescence methods can be used to determine Matrix Metalloproteinase-7 localization. The obtained tissues were compared and evaluated semi-quantitatively by taking photographs in appropriate areas. Morphological comparison between control and experimental groups revealed a marked expansion in the uterine lumen across control categories, along with increased epithelial cell diversity and enhanced delineation of epithelial cell borders. Significant differences in Matrix Metalloproteinase-7 expression were observed between groups. In conclusion, follitropin alfa alters the hormonal milieu of uterine tissue, leading to matrix-related structural changes.

Ratlarda Uterusta Follitropin Alfanın Matriks Metalloproteinaz-7 Ekspresyonuna Etkisi

Öz

Follitropin alfa, gonadotropinler olarak tanımlanan hormon ailesine ait bir çeşit folikül uyarıcı hormondur ve yardımla üreme teknolojisinde kullanılmaktadır. Çalışmamızda, Follitropin alfa verilmiş hayvanların uterus dokusu üzerinde, Matrix Metalloproteinaz-7 ekspresyonunun gösterilmesi amaçlanmıştır. Çalışmamızda, vücut ağırlıkları ortalama 200-220 gram ağırlığında 8-10 haftalık Wistar albino cinsi 16 adet erişkin dişi rat kullanıldı. Diöstrus dönemini 3’e bölecek şekilde Follitropin alfa (4 IU) enjeksiyonu yapıldı. 21 günün sonunda uterus dokuları alındı. Dokudan alınan örnekler %10 tamponlanmış nötral formalin çözeltisinde tespit edildi. 3 µm kalınlığında kesitler alınan doku uterus örneklerine genel morfoloji için Periyodik asit–Schiff ve Hematoksilen-eozin boyama yöntemleri, Matriks Metalloproteinaz-7 lokalizasyonun belirlenebilmesi için immünfloresan yöntemler uygulandı. Elde edilen dokular karşılaştırılarak değerlendirildi ve uygun alanlardan fotoğraflar çekilerek semikantitatif olarak değerlendirdi. Kontrol ve deney grupları morfolojik olarak karşılıklı değerlendirildiğinde, deneysel grupta uterus lümeninin kontrol grubuna oranla çok fazla genişlediği, epitel tabakasında yer alan hücrelerin çekirdek sayılarında artışla beraber epitel tabasının da hücre sınırlarında artış meydana geldiği gözlendi. Matrix Metalloproteinaz-7 ekspresyonunda ise gruplar arası yine anlamlı farklılıklar izlendi. Sonuç olarak Follitropin alfa’nın uterus dokusu üzerinde hormonal düzenlenim açısından farklılıklara ve matriks bozulması gibi değişikliklere sebep olabileceği düşünülmektedir.
