In-silico investigation of structural, electronic and biological properties of 2-[5-(4-chlorophenyl)-1H-pyrazole-3-yl]-6-(2-hydroxymethyl-1-methylpyrrolidine-3-yl)-3,5-dimethoxy-phenol compound

Öz

Pyrazole derivatives occupy an important position in medicinal and pharmaceutical research owing to their diverse biological properties. Among these activities, anti-inflammatory and analgesic effects have attracted considerable attention because pyrazole-based compounds are known to interact with important mediators involved in inflammatory pain pathways. However, studies systematically investigating the relationship between molecular structure and the physicochemical/electronic properties of these compounds remain limited. In the present study, the structural, electronic, and biological properties of 2-[5-(4-chlorophenyl)-1H-pyrazole-3-yl]-6-(2-hydroxymethyl-1-methylpyrrolidine-3-yl)-3,5-dimethoxyphenol (HMP2) were comprehensively examined using in silico approaches. The most stable molecular geometry was optimized at the second-order Møller–Plesset perturbation theory (MP2) level, and structural stability was evaluated through bond lengths and bond angles. Chemical reactivity and kinetic stability were assessed using HOMO–LUMO energy gap calculations, while electrophilic and nucleophilic regions were identified through molecular electrostatic potential (MEP) maps. Intramolecular charge-transfer behavior was investigated using nonlinear optical (NLO) properties, Mulliken atomic charge distributions, and Natural Bond Orbital (NBO) analyses. Furthermore, molecular docking studies against inflammation-related target proteins revealed that HMP2 exhibits favorable binding modes and strong binding affinities, indicating its potential anti-inflammatory activity. ADMET predictions further demonstrated that the compound possesses acceptable pharmacokinetic and toxicological characteristics. These findings suggest that HMP2 may be considered a promising candidate for the development of novel anti-inflammatory agents.
