Tunceli dağ sarımsağı (Allium tuncelianum) farklı eksraksiyonlarında LC-MS/MS ile fenolik bileşik miktarlarının karşılaştırılması

Öz

Bu çalışmada ekstraksiyon yöntemi ve çözgen farklılığının, Tunceli Dağ Sarımsağı'nda fenolik bileşen karekterizasyonuna etkisinin araştırılması amaçlanmıştır. Fenolik bileşiklerin karakterizasyonu için; 25 fenolik bileşik kullanılarak, yeni bir LC-MS/MS metodu geliştirilmiş ve bu metod Tunceli Dağ Sarımsağı (TDS) için kullanılmıştır. Ekstraksiyon yöntemi olarak; infüzyon ve hidrolizasyon metodu, çözgen olarak; etanol, metanol, su ve etanol/su (% 50 [v/v]) olmak üzere 4 farklı çözücü kullanılmıştır. TDS içerisinde 25 farklı fenolik bileşen aranmış ve bunlardan 16 fenolik bileşenin var olduğu tespit edilmiştir. TDS’nin major komponentlerinin; pirokatekol (14018.98±3.61 µg L-1), vanilik asit (1887.24±0.397 µg L-1) ve fumarik asit (1812.34±0.211 µg L-1) olduğu belirlenmiştir. TDS nin fenolik içeriğinin çeşitlilik açısından zengin, ancak miktar açısından fakir olduğu görülmüştür. Genel olarak en iyi çözgenin etanol ve en ideal ekstraksiyon yönteminin ise hidrolizasyon olduğu görülmüştür. Sonuç olarak, farklı analiz tekniği, farklı ekstraksiyon işlemi ve farklı çözgen kullanımının; Allium tuncelianum için, fitokimyasal içerik üzerine oldukça önemli etkilerinin olduğu anlaşılmıştır.

Comparison of amount of phenolic compounds by using LC-MS / MS in Tunceli mountain garlic (Allium tuncelianum) different extracts

Öz

In this study, it was aimed to investigate the effect of extraction methods and solvent differences on phenolic component characterization in Tunceli Mountain Garlic. For the characterization of phenolic compounds; A new LC-MS / MS method was developed using 25 phenolic compounds and this method was used for Tunceli Mountain Garlic (TDS). As extraction method; infusion and hydrolization method, as solvent; ethanol, methanol, water and ethanol / water (50 % [v/v]), including four different solvents were used. In TDS, 25 different phenolic components were searched and 16 phenolic components were found out of them. Major components of TDS; pyrocatechol (14018.98 ± 3.61 µg L-1), vanicic acid (1887.24 ± 0.397 µg L-1) and fumaric acid (1812.34 ± 0.211 µg L-1). The phenolic content of TDS was found to be rich in diversity but poor in quantity. In general, the best solvent was ethanol and the most ideal extraction method was hydrolization. As a result, different analysis technique, different extraction process and different solvent use; For Allium tuncelianum, it was found to have significant effects on phytochemical content.
