Polyporus squamosus (Huds.) Fr. OBCC 5003 Suşu ile Biyokütle Üretimi Üzerine Çalışmalar

Öz

Bu çalışmada, Fındıksuyu Köyü’ nden (Geyve-Sakarya) toplanarak dikaryotik misel formunda izole edilen Polyporus squamosus OBCC 5003 suşunun misel gelişimi üzerine farklı sıcaklık ve pH değerleri ile besiyeri içeriğinin etkileri incelenmiştir. Araştırma sonucunda 8 farklı inkübasyon sıcaklığı (10–45 °C) ve 9 farklı pH derecesi (4.0-8.0) arasından en iyi misel gelişimi gözlenen sıcaklık ve pH değerleri 30 °C ve 5 olarak belirlenmiştir.  Besin tercihinin belirlenmesi amacı ile P. squamosus OBCC 5003 suşu 14 farklı besiyerinde büyütülerek günlük misel gelişim değerleri (koloni çapı ve büyüme hızı) belirlenmiştir. En yüksek gelişim hızı, 35,48 mm/gün değeri ile glukoz malt yeast agar besin ortamında belirlenmiştir. Elde edilen veriler P. squamosus OBCC 5003 suşunun biyoreaktör sisteminde derin kültür yöntemi ile büyütülmesi amacı ile kullanılmış ve organizmanın büyüme parametreleri (biyokütle üretimi ve substrat tüketimi) belirlenmiştir.
