Drying Methods Used for Preservation of Microbial Cultures

Abstract

All laboratories that do microbiological researches need culture collections because laboratories have to form their own collections and preserve on suitable conditions them for control of testing. Basic principle of culture preservation is to keep cells alive and in pure form for a long time without causing mutations and variations in microorganisms. Drying is the oldest method of culture preservation. Because it is an inexpensive and easy method, it is frequently used for the preservation of microbial cultures. Stages of all drying methods are generally similar to each other. Initially, culture is mixed with protectants and dried with a desired process. Then, dry culture is stored. When needed, cells are activated by rehydrating culture. This review explains simple drying methods like sand drying, drying on paper disc, drying with gelatin and silica gel, self-drying and alternative drying methods except lyophilization like spray drying, fluidised bed drying, foam drying and drying under vacuum

Mikroorganizma Kültürlerinin Korunmasında Kullanılan Kurutma Yöntemleri

Abstract

Mikrobiyoloji alanında çalışma yapan tüm laboratuvarlar kültür koleksiyonlarına ihtiyaç duymaktadır. Çünkü yapılan testlerin kontrolü amacıyla laboratuvarların, kullandıkları kültürlerin koleksiyonlarını oluşturması ve bu kültürleri uygun koşullarda muhafaza etmeleri gerekmektedir. Kültür korumanın temel prensibi, varyasyon veya mutasyona uğratmadan mikroorganizmayı saf halde ve uzun süre canlı tutmaktır. Kurutma yöntemi bilinen en eski kültür muhafaza yöntemidir. Ucuz ve kolay bir yöntem olduğundan pek çok mikroorganizma kültürünün muhafazasında sıklıkla kullanılmaktadır. Tüm kurutma yöntemlerinde aşamalar genel olarak birbirine benzemektedir. Öncelikle mikroorganizma kültürü koruyucu maddelerle karıştırılır ve istenilen yöntemle kurutulur. Ardından kurutulan kültür muhafazaya alınır. İhtiyaç duyulduğunda ise kültür rehidre edilerek hücreler aktifleştirilir. Bu derleme, kumda kurutma, kâğıt disk üzerinde kurutma, jelatin ve silikajelde kurutma ile kendini kurutma self-drying gibi basit kurutma yöntemlerinin yanı sıra sprey kurutma, akışkan yatak kurutucularda kurutma, köpük kurutma ve vakumda kurutma gibi liyofilizasyon dışındaki son dönemde ön plana çıkan kurutma yöntemlerini ve kullanım alanlarını irdelemek amacıyla hazırlanmıştır
