Green Synthesis of Silver Nanoparticles Mediated by Royal Jelly: Structural Characterization and Antioxidant Capacity

Abstract

The green synthesis of silver nanoparticles (AgNPs) using natural biomolecules has attracted increasing attention due to its environmental sustainability and enhanced biological functionality. In this study, royal jelly (RJ) was employed as both a reducing and stabilizing agent for the green synthesis of AgNPs, and the structural properties and antioxidant capacity of the obtained nanoparticles were systematically evaluated. The formation of RJ-mediated AgNPs was confirmed by the characteristic surface plasmon resonance band observed in UV–Vis spectroscopy. Transmission electron microscopy analyses revealed the formation of predominantly spherical nanoparticles with a nanoscale size distribution, surrounded by an organic surface coating. Fourier transform infrared (FTIR) spectroscopy demonstrated that RJ -derived proteins and phenolic compounds play a significant role in nanoparticle reduction and stabilization. The antioxidant capacity of RJ-mediated AgNPs was evaluated using the DPPH free radical scavenging assay and compared with that of RJ. The results showed that RJ–AgNPs exhibited significantly higher free radical scavenging capacity than RJ alone, particularly at higher concentrations. In contrast, total phenolic content analysis revealed no statistically significant difference between the RJ and RJ–AgNP groups. This finding suggests that the enhanced antioxidant capacity is not directly related to an increase in phenolic content but rather to the immobilization of phenolic compounds on the nanoparticle surface and the increased effective surface area provided by the silver nanoparticles. Consequently, RJ-mediated AgNPs can be considered biofunctional nanomaterials with promising potential for biomedical and pharmaceutical applications.
