Pichia pastoris SMD1168 Suşu Kullanılarak Rekombinant Ekspanzin Üretimi

Öz

Bu çalışmanın
amacı son yıllarda rekombinant protein üretiminde başarılı bir şekilde
kullanılan metilotropik
P. pastoris SMD1168 suşunu kullanarak domates
bitkisinden izole edilen ekspanzin enziminin üretilmesidir. Bu amaçla domates (
Solanum
lycopersicum)
genomundan toplam RNA izole edilmiş ve spesifik primerler
vasıtasıyla çoğaltılarak pHO1 sinyal sekans bölgesine sahip pHIL-S1 vektörüne
aktarılmıştır. PZR yöntemi ile 918 baz çiftlik LeExp1 geninin vektör
içerisindeki varlığı tespit edilmiş ve hedef geni taşıyan pHIL-S1 vektörü
çoğaltılarak
Pichia pastoris SMD1168
suşuna elektroporasyon yolu ile transfer edilmiştir. Transgenik
P. pastoris SMD1168 suşundan ekspresyon
yolu ile ekspanzin proteini üretilmiş ve ekspresyon sonunda toplam protein
konsantrasyonu ile kuru hücre ağırlığı ölçümleri yapılmıştır. SDS-PAGE analizi
ile elde edilen proteinin ekspanzin olduğu doğrulanmıştır. Bu çalışmanın sonucunda
Pichia pastoris SMD1168  suşu ve pHIL-S1 vektörü kullanılarak
rekombinant ekspanzin proteini üretimi gerçekleştirilmiştir.
