Correlation Between Color Parameters (CIE Lab*), Phenolic and Flavonoid Contents, and Antioxidant Capacity of Ethanolic Propolis Samples

Abstract

Raw propolis is a natural product obtained directly from beehives and widely used for its bioactive properties. In this study, ten raw propolis samples collected from different regions of Türkiye were analyzed in terms of color parameters (CIE Lab*), total phenolic content (TPC), total flavonoid content (TFC), antioxidant capacity (FRAP), DPPH radical scavenging activity, and phenolic composition using RP-HPLC-PDA. The relationships between color characteristics, phenolic composition, and antioxidant activity were statistically evaluated. TPC values ranged from 12.95 to 224.45 mg GAE/g, showing significant variability among samples. A strong positive correlation was observed between phenolic content and antioxidant capacity. Major phenolic compounds commonly detected across the samples included caffeic acid, chrysin, pinocembrin, and p-coumaric acid, all of which were present in every sample, whereas caffeic acid phenethyl ester (CAPE) was present in all samples except PS-7. The results demonstrate that phenolic composition plays a key role in determining the antioxidant capacity and overall quality of propolis.
