Chemical characterization and antimicrobial–antimutagenic activities of propolis samples collected from Duzce city center

Abstract

Propolis is a natural product produced by bees from plant resins, possessing a rich chemical composition and various biological activities. This study aimed to chemically characterize propolis samples obtained from Düzce city center and determine their antibacterial, mutagenic, and antimutagenic activities. Seventeen different component analyses were performed on ethanol-based and water-based extracts prepared from propolis samples. The highest concentration was found to be caffeic acid phenethyl ester at 11482.18 µg/mL in ethanol-based propolis extract and caffeic acid at 726.85 µg/mL in water-based propolis extract. Antimicrobial activity was evaluated against 14 different test microorganisms (eight Gram-negative and six Gram-positive bacteria) using the agar well diffusion method. MIC values for ethanol-based propolis extract ranged from 0.18 to 23.60 mg/mL, and MIC values for water-based propolis extract ranged from 0.598 to 23.90 mg/mL. Specifically, it was determined that ethanol-based propolis extract exhibited a stronger inhibitory effect on Gram-positive bacteria at lower concentrations compared to water-based propolis extract. Mutagenicity and antimutagenicity analyses were performed using the Allium cepa test. At the tested concentrations, propolis extracts were found not to exhibit mutagenic effects. In antimutagenicity analyses, it was determined that propolis extracts showed a protective effect against H₂O₂-induced DNA damage, and significantly reduced damage, especially at low concentrations (2.99-0.18 mg/mL). The results indicate that Düzce propolis possesses significant antibacterial and genoprotective properties.
