Effects of Zinc and Activated Carbon Nanoparticles on the Motility of Ram Spermatozoa during Storage at 4°C

Abstract

This study evaluated the effects of zinc and activated carbon nanoparticles, added at different concentrations to a Tris-based extender, on the motility of ram semen during liquid storage at +4°C. Semen was collected from four fertile Kangal-breed rams across four replicates, pooled, and divided into seven groups: control, ZnNP50, ZnNP100, ZnNP200, CNP50, CNP100, and CNP200. Total motility was assessed every 8 hours until complete loss, and data were analyzed using mixed analysis of variance with the Šídák test and Kaplan-Meier survival analysis with the log-rank test. Motility loss in the control group occurred between 136 and 160 hours. No nanoparticle group differed significantly from the control group according to the Šídák test, although the CNP50 and CNP100 groups showed significantly lower motility than the ZnNP50 and ZnNP200 groups during mid-to-late storage. Survival analysis showed that only the CNP200 group remained motile significantly longer than the control group after correction for multiple comparisons (median 168 versus 136 hours). These findings suggest that activated carbon nanoparticles, particularly at 200 µg/ml, may support motility persistence during the liquid storage of ram semen, warranting further mechanistic investigation.
