Monitoring Changes in Citrus Chlorotic Dwarf-associated Virus Titer under Natural Field Conditions

Abstract

The citrus chlorotic dwarf-associated virus (CCDaV), which was first detected in citrus grown in Turkey, is one of recently emerged disease of citrus. The molecular diagnostic and characterization of the virus was carried out in recent years. However, the information available on the etiology of the virus is extremely limited. This study aimed to determine the presence of CCDaV in different tissues, and seasonal and vertical changes of virus titter in terms of the etiology of the causal agent. The presence and changes in CCDaV titter were investigated from samples taken from different tissues and parts of a naturally infected with lemon tree throughout the four seasons. As a result of conventional and real-time PCR tests, CCDaV was detected in leaves, flowers and fruits. Analysis of titter changes of CCDaV in different seasons and directions revealed that the virus titter was statistically at the highest level in the northward direction in the spring months. However, it has been determined that routine CCDaV surveys can be performed from leaf samples at any direction of tree and time of the year with conventional PCR as well.
